Do I have to get Manual accepted by the AIA before implementing the program in place for the demonstration of Mock-up?

Can I use prepared Manual and perform my purchasing activities, Design report preparation and ITP development for review and hold point by ANI.

Yes. Ideally, the manual has to be accepted by ANIS before implementing the same.

If the demonstration of mock-up is for the ASME Nuclear survey for Nuclear Certificate of Authorization then your QA Manual has already accepted by your AIA.

Basically the correct sequence is

Prepare the manual
approve it
get it accepted by ANIS
distribute it
training and indoctrination
design
qualification of vendors
purchasing, etc.

If during the preparation for the Survey a PO is issued to a vendor not qualified, it is a violation of the Code and QA manual.
If fabrication is started prior to ANI acceptance of traveler, I recommend to cancel the Survey. It would be a waste of time and money.
